HPLC Preparative Equipment
The NACC is equipped with a diverse range of HPLC instruments, offering various purification capabilities across different scales and chromatographic techniques. These include ion exchange (IEX), reverse phase (RP), and ion-pair reverse phase (IP-RP), allowing precise separation and purification of complex oligonucleotides.
Two Agilent 1290 Infinity II Preparative LC systems support high-throughput, UV-based preparative purification. These systems feature open-access automated sample injection and fraction collection, with 15 mL and 90 mL injection loops respectively. They accommodate flow rates of up to 200 mL/min and operate at a maximum pressure of 600 bar.
Two Agilent 1260 Infinity II UV-based Preparative LC systems are used for routine purification workflows. These systems offer flow rates of up to 100 mL/min and a maximum operating pressure of 600 bar. External pumps are used for sample injections to accommodate large-volume samples.
Two Agilent 1200 and 1260 semi-preparative UV-based systems are used for smaller-scale purifications below 1 µmol, supporting high-precision workflows.
These systems collectively support the NACC’s mission to provide high-quality purification of therapeutic oligonucleotides at scales ranging from nanomoles to grams.
