HPLC Chromatography
For synthetic peptides, reverse-phase HPLC is performed on the entire batch of material obtained from the synthesizer instrument. Preparative scale (up to 200 mg) reverse-phase HPLC is routinely performed using acetonitrile gradients and Water's equipment with variable wavelength spectrophotometric detection.
Sep-Pak Purification
For 70% purity, the peptide can be Sep-Pak purified and then lyophilized.
Amino Acid Analysis
Amino acid analysis involves pre-column derivitization with Accu-Tag reagent, followed by automated HPLC separation and computerized peak integration using the Water's Millenium system.
Peptide Conjugation
Peptides are routinely conjugated to proteins to prepare immunogens with incorporations of up to 1000 mol/mol. Generally, this involves the use of enriched forms of KLH or BSA and coupling reagents that specifically crosslink Lys or Tyr residues, followed by dialysis and lyophilization.
